Testing Algorithm:

  • BCR-ABL1 Reflex to ABL1: ABL1 Kinase Domain Mutation will be run when BCR-ABL1 is positive.
  • JAK2 V617 Reflex to JAK2 Exon 12-14: Exon 12-14 will be run when V617F result is negative.
  • MPN Standard Reflex Panel: JAK2 V617F is run first. JAK2 Exon 12-14 will be run when V617F is negative. MPL will be run when JAK2 Exon 12-14 is negative.

Specimen Requirements

Specimen Type: Bone Marrow Aspirate

Container/Tube: EDTA (lavender top)

Specimen Volume: 2 mL

Collection Instructions:

  1. Invert several times to mix bone marrow.
  2. Send specimen at ambient temperature in original container.
  3. Include a YPI requisition.

Specimen Type: Peripheral Blood

Container/Tube: EDTA (lavender top)

Specimen Volume: 5 mL

Collection Instructions:

  1. Invert several times to mix blood.
  2. Send specimen at ambient temperature in original container.
  3. Include a YPI requisition.

Specimen Type: Fresh Tissue

Container/Tube: RPMI

Specimen Minimum Size: 2 pieces of tissue 0.2 cm3

Collection Instructions: 

  1. Collect tissue and place in RPMI.  Frozen tissue is also acceptable.
  2. Send RPMI specimen at ambient temperature.
  3. Include a YPI requisition.

 

Specimen Type: FFPE lymph node in block

Container/Tube: Block cassette & 1 H&E slide

Collection Instructions: 

  1. Send specimen at ambient temperature.
  2. Include a YPI requisition.

 

Specimen Type: FFPE lymph node cut slides

Container/Tube: 1 H&E slide plus 5-10 positively charged unstained slides cut at 5 or more microns.

Collection Instructions: 

  1. Do not use zinc fixatives.
  2. Send specimen at ambient temperature.
  3. Include a YPI requisition.

 

Specimen Rejection:

1.  Unlabeled specimens.

2.  Specimens collected in wrong container/tube.

3.  Specimens retained past viability.

4.  Specimens processed with zinc fixatives.
